چکیده

The nerve fiber distribution in the epidermis of the hairless rat skin was studied light microscopically by means of zinc iodide-osmium tetroxide staining. Two different morphological types of free nerve fiber endings could be detected: clusters of relatively thick nerve fibers stretched up through the spinous layer up to the granular layer sending off terminal branches. In addition, many solitary thin varicose nerve fibers were seen within the epidermis. The observed discrepancies in nerve fiber diameters appeared to be larger than those reported for human intraepidermal nerve fibers in recent immunohistochemical studies. Moreover, dendritic cells, most probably representing Langerhans cells, could be selectively stained. These cells appeared to be in a close location to thin varicose nerve fibers. Both types of demonstrated free nerve endings have to be functionally connected with different sensoric functions. Possibly, a subpopulation of the thin nerve fibers might possess primarily a nociceptive task, whereas the thick ones have most probably to be regarded as mechanoreceptive. The nerve fibers innervating dendritic cells appear to be identical to the peptidergic ones which may regulate the antigen-presenting capacity of these cells. Due to its selectivity for intraepidermal nerve fibers, the used method might supplement immunohistochemical procedures in a helpful manner.

Investigation of the ultrastructurel of different cells in lymph nodules by using zinc iodide osmium tetroxide technique.

The zinc iodide -osmium tetroxide (ZIO) fixation/staining method was applied for neurocytological study and also to examine several other tissue samples including human blood and bone marrow on nerve endings in the median eminence, epidermal langerhans cells of lymphoid tissue. Although precise specificity can not be attributed to the staining reaction. Interesting staining patterns for differe...

Enhancement of the Granulation of Adrenergic Storage Vesicles in Drug-free Solution

Small granular vesicles within autonomic nerve terminals are now generally identified as storage sites for noradrenalin (Hökfelt, 1968) . However, these granules do not always appear in the one preparation after different fixation procedures, and even with the same fixation technique they can occur in some adrenergically innervated tissues but not in others. It has been shown by Richardson (196...

Aspects of turnover and biogenesis of synaptic vesicles at locust neuromuscular junctions as revealed by zinc iodide-osmium tetroxide (ZIO) reacting with intravesicular SH-groups

Retractor unguis nerve muscle preparations from the locust were subjected to the zinc iodide-osmium tetroxide reaction (ZIO) after pre-fixation in glutaraldehyde. Applied for 18 h at 4 degrees C in the dark, ZIO reacts at pH 4.2--4.0 fairly selectively with the matrix of synaptic vesicles. Approximately 53% of the vesicles are completely and 4% partially stained. The percentage of ZIO-positive ...

Osmium Zinc Iodide Reactive Sites in the Epidermal Langerhans Cell

Fixation of epidermis with a mixture of osmium tetroxide and zinc iodide (OsO(4)-ZnI(2)) for 24 hr renders the central periodic lamella of the Langerhans cell granule (LCG), the Golgi region, and the nuclear envelope of epidermal Langerhans cells preferentially visible. The use of this technique on Langerhans cells in normal epidermis and in epidermis of patients with histiocytosis (Letterer-Si...
